QuantStudioTM Absolute QTM Digital PCR System (ThermoFisher Scientific)

  • Quantitative multiplex assay: quantification of mutations at codons D835 and I836, located in the TKD2 domain of FLT3, in genomic DNA from peripheral blood by means of digital PCR (dPCR).
  • FLT3-TKD2 dPCR OncoKit is intended for use in research and is aimed exclusively at healthcare workers and professionals working in molecular biology.

FLT3-TKD2 dPCR OncoKitis designed to quantify pathogenic variant at codons D835 and I836 of the second tyrosine kinase domain (TKD2) of the FLT3 gene. The kit utilizes digital PCR (dPCR), a highly sensitive, precise, and accurate end-point PCR method that allows absolute quantification of a sample that is subdivided into 20,000 partitions.

The kit employs a combination of oligonucleotides and fluorescent hydrolysis probes in a non-competitive multiplex assay for quantification of the alternative allele.

  • Sample type: Genomic DNA from peripheral blood
  • Positive control: Synthetic DNA with heterozygous FLT3 p.D835Y
  • Necessary amount of DNA: 65 ng.
  • Limit of detection: 0,2% ± 5,4E-4.
  • Limit of quantification: 0,5%
  • Number of reactions per sample: 1
  • Number of targets: 2 (altered allele and unchanged, wild-type, allele of FLT3).
  • Duration of the dPCR program: 1 h 45 min.
