Consiste numa análise por PCR que permite amplificar e, simultaneamente, quantificar o produto dessa PCR. Para tal, baseia-se no princípio da PCR convencional, mas utiliza um fluoróforo que permite medir a quantidade de produto amplificado em tempo real.

Em função da sua dimensão e das regiões de interesse, podemos oferecer uma abordagem baseada em sequenciação Sanger ou em NGS (enriquecimento por amplicões ou por sondas de hibridação). A abordagem baseada em NGS permite a deteção de variações no número de cópias (CNV).

Técnica semiquantitativa e amplamente validada nos laboratórios de genética molecular que permite o diagnóstico de patologias devidas a variantes no número de cópias e, em alguns casos, a alterações de metilação. Existem numerosos kits comerciais para o estudo de genes individuais, painéis de genes relacionados com patologias específicas ou regiões cromossómicas extensas envolvidas em síndromes de microdeleção/microduplicação. A HIC presta serviços de MLPA baseados nos kits da MRC-Holland.
